An international medical study has yielded new findings that are of important reference value for disease prevention. Previous studies have shown that heat-resistant protein 90 ((Hsp90) inhibitors can reduce seizures of temporal lobe epilepsy (TLE) by promoting the proliferation of excitatory amino acid transfer 2 ((EAAT2), or GLT-1). Although Hsp90 inhibitor 17-AGG is effective, concerns have been raised about its long-term toxicity.

The aim of this study is to target the extract of anamycin derivatives with higher absorption and lower toxicity by screening them. Reblastatin emerged as the most likely candidate in the selection of nine naturally occurring derivatives and their derivatives. It demonstrated significant cellular toxicity in HEK293 and HepG2 cells due to its high affinity for Hsp90.

Similar to 17-AGA, Reblastatin achieves high affinity for Hsp90 by disrupting the association of Hsp90, EAAT2, 20S protein enzymes and by inhibiting the interaction between Hsp90, EAAT2, 20S protein enzymes and 20S protein enzymes, thus demonstrating significant cytotoxicity in HEK293 and HepG2 cells. In the carnic acid-induced TLE mouse model, reactivated proteins (reblastatin) decreased the frequency of seizures by 50% and long-term treatment showed toxicity comparable to vehicle control. Furthermore, behavioral tests showed the neuroprotective role of reactivated proteins in mice.
