Disordered metabolism and the potential impact of carcinogens on the development of cancer

An international medical study has brought new findings and related findings have important reference value for disease prevention. Endocrine disorders (EDCs) constitute a group of heterogeneous chemicals that interfere with endocrine regulation and thus affect the initiation and progression of cancer risk. Their mechanisms of action include interaction with endocrine receptors, regulation of endocrine synthesis and metabolism, changes in DNA epigenetics, and activation of specific signaling pathways.

In recent years, the AOP framework has been significantly enhanced because it is able to link molecular events triggered by molecular initiation events influenced by ECs to clinical outcomes, including cancer mutations. Thyroid hormone plays a multifaceted role in regulating cell division, differentiation, and metabolism, while at multiple levels the dysregulation of thyroid hormone influenced by ECs may affect the development of several types of cancer. The purpose of this paper is to summarize the latest knowledge of the AOP framework on the major mechanisms of induced endocrine disorders and their potential effects on the initiation and progression of cancer and predict the risks associated with endocrine disorders.
